Bepreve Eye Drops Bepotastine besilate 1,5% ophthalmic solution is an antagonist of H1-receptors, the drug is approved for the topical treatment of itching associated with allergic conjunctivitis in patients of 2 years and older. Allergic conjunctivitis is the most common form of ocular allergy, with itching as a primary symptom. Treatment of ocular itching include antihistamines (oral and ophthalmic), ophthalmic drugs stabilize mast cell membrane, non-steroidal anti-inflammatory drugs (NSAIDs), and, in severe cases, corticosteroids. Ocular antihistamines are effective and begin to act more quickly than oral antihistamines. Bepotastine was first developed in Japan as an oral systemic drug for the treatment of allergic rhinitis.
Two randomized double-blind, placebo-controlled study of 237 patients with allergic conjunctivitis in the history showed that the ophthalmic solution bepotastine 1.5% more effective than placebo in reducing ocular itching intensity after 15 minutes and 8 hours.
Pharmacology and Pharmacokinetics of Bepreve Eye Drops
Bepotastine is similar in structure to antihistamines belonging to the group of ethanolamines (Figure 2) has a local activity, is a direct antagonist of the H1-receptor and an inhibitor of the release of histamine from mast cells. The drug takes effect after about 3 minutes after application, and duration of effect is 8 hours. Vepotastine minimally absorbed when applied topically and is excreted by the kidneys in unchanged form.

In clinical studies, the most common adverse reactions in the application of bepotastine were marked by a slight taste disturbance, registered at about 25% of patients. Other side effects include eye irritation, headache, and nasopharyngitis (2% -5%). During the 6-week safety study, which included 127 children aged ≥ 3 years, no serious side effects were observed. Bepotastine has terratogennym action and belongs to the category C drug and should be used with caution during pregnancy and lactation. Given the topical drug is minimally absorbed, and do not interact with cytochrome (CYP3A4, CYP2C9, CYP2C19), respectively, significant drug interactions are expected.
